Un ensayo multicéntrico en fase II ha evaluado lurbinectedina, un inhibidor selectivo de PharmaMar de transcripción activada de ciertos genes, en pacientes con cáncer de mama metastásico que tengan mutaciones en los genes BRCA 1 y/o BRCA 2.

Zepsyre® (lurbinectedina), de Pharma Mar, muestra una actividad clínica significativa en los pacientes con cáncer de mama metastásico con mutaciones en los genes BRCA1 y/o BRCA2. Esta es la conclusión que recogen las revistas científicas The Lancet Oncology y Journal of Clinical Oncology, en sus números de septiembre y noviembre, respectivamente, las cuales han publicado los resultados positivos del estudio de PharmaMar de Fase II.

Este estudio evaluó la actividad clínica de lurbinectedina en pacientes con cáncer de mama metastásico que presentaban mutaciones en los genes BRCA1 y/o BRCA2.

Es importante destacar que en pacientes con mutaciones en BRCA2, la tasa global de respuestas confirmadas fue del 61%, la supervivencia media libre de progresión fue de 5,9 meses y la mediana de la supervivencia global fue de 26,6 meses.

En la población no tratada previamente con inhibidores de PARP, la tasa de respuestas sube a un 72%, que es la tasa de respuestas más alta publicada hasta hoy en esta indicación.

Las 54 pacientes de cáncer de mama con mutaciones en los genes BRCA 1 y/o BRCA 2 fueron reclutados de 11 centros de investigación en Estados Unidos y en España.

"Lurbinectedina es un inhibidor selectivo de la transcripción activa de genes que codifican proteínas", explica Judith Balmaña (Instituto de Oncología Vall d'Hebron) en The Lancet Oncology. "En este ensayo de fase II, lurbinectedina mostró una eficacia notable en pacientes con cáncer de mama metastásico y mutación de la línea germinal BRCA1/2".
